SHANGHAIEncorafenib (Braftovi®), a potent, highly selective BRAF kinase inhibitor approved in China in June 2025, is now officially available at Jiahui International Cancer Center. Indicated in combination with cetuximab for adult patients with BRAF V600E-mutated metastatic colorectal cancer (mCRC) after prior systemic therapy, this regimen brings a new precision-medicine option to one of the most challenging colorectal cancer subtypes.

A Precision Breakthrough for BRAF V600E+ Metastatic Colorectal Cancer

Colorectal cancer is one of the most common malignancies in China. Approximately 7% of patients carry the BRAF V600E mutation. These patients typically respond poorly to conventional chemotherapy, experience rapid disease progression, and have limited treatment options, posing a significant survival challenge.

The encorafenib plus cetuximab combination precisely targets BRAF V600E-mutated mCRC, particularly as a later-line option after prior treatment failure. By potently inhibiting mutant BRAF protein activity and blocking the MAPK signaling pathway, encorafenib effectively suppresses tumor growth and spread, providing a clear targeted treatment pathway for this high-risk subgroup.

Improved Response Rates and Durable Remission

Encorafenib demonstrated outstanding efficacy in the pivotal Phase III BREAKWATER study:

  • Objective Response Rate (ORR): 60.9%, significantly higher than the 40.0% seen with traditional chemotherapy plus bevacizumab.
  • Median Duration of Response (mDoR): 13.9 months, longer than the 11.1 months in the control group, suggesting more durable treatment response.
  • Favorable Safety Profile: The most common adverse reactions were fatigue and arthralgia, mostly mild to moderate, with good patient tolerability.
